Proposed top-level domain

The Domain Name System of the Internet consists of a set of top-level domains which constitute the root domain of the hierarchical name space and database. In the growth of the Internet, it became desirable to expand the set of initially six generic top-level domains in 1984. As a result new top-level domain names have been proposed for implementation by ICANN. Such proposals included a variety of models ranging from adoption of policies for unrestricted gTLDs that could be registered by anyone for any purpose, to chartered gTLDs for specialized uses by specialized organizations. In October 2000, ICANN published a list of proposals for top-level domain strings it had received.

MED28

Mediator of RNA polymerase II transcription subunit 28 is an enzyme that in humans is encoded by the MED28 gene. It forms part of the Mediator complex.

Function

Subunit Med28 of the Mediator may function as a scaffolding protein within Mediator by maintaining the stability of a submodule within the head module, and components of this submodule act together in a gene-regulatory programme to suppress smooth muscle cell differentiation. Thus, mammalian Mediator subunit Med28 functions as a repressor of smooth muscle-cell differentiation, which could have implications for disorders associated with abnormalities in smooth muscle cell growth and differentiation, including atherosclerosis, asthma, hypertension, and smooth muscle tumours.

MED12

Mediator of RNA polymerase II transcription, subunit 12 homolog (S. cerevisiae), also known as MED12, is a human gene found on the X chromosome.

Clinical significance

Mutations in MED12 are responsible for at least two different forms of X-linked dominant mental retardation, Lujan-Fryns syndrome and FG syndrome, as well as instances of prostate cancer.

Mutations in MED12 are associated to uterine leiomyomas and breast fibroepithelial tumors (e.g. fibroadenoma and phyllodes tumors).

MED13

Mediator complex subunit 13 is a protein that in humans is encoded by the MED13 gene.

Function

This gene encodes a component of the mediator complex (also known as TRAP, SMCC, DRIP, or ARC), a transcriptional coactivator complex thought to be required for the expression of almost all genes. The mediator complex is recruited by transcriptional activators or nuclear receptors to induce gene expression, possibly by interacting with RNA polymerase II and promoting the formation of a transcriptional pre-initiation complex. The product of this gene is proposed to form a sub-complex with MED12, cyclin C, and CDK8 that can negatively regulate transactivation by mediator.

MED29

Mediator of RNA polymerase II transcription subunit 29 is a transcription suppressor that in humans is encoded by the MED29 gene. It represents subunit MED29 of the Mediator complex.

MED29, along with Med11 and MED28 in mammals, is part of the core head-region of the Mediator complex. Med29 is the apparent orthologue of the Drosophila melanogaster Intersex protein (IXL), which interacts directly with, and functions as a transcriptional coactivator for, the DNA-binding transcription factor Doublesex, so it is likely that mammalian Med29 serves as a target for one or more DNA-binding transcriptional activators.
